Effects of Citric Acid Treated Root Surfaces on Attachment and Proliferation of Gingival Fibroblasts

Abstract
This invitro study was undertaken to observe whether citric acid application aids attachment and proliferation of fibroblasts to the root surfaces of periodontally diseased teeth. 15 normal teeth as normal group, which were extracted on orthodontic purpose, were selected and were treated to remove the periodontal ligament and soft tissue. 30 diseased teeth, which were considered as hopeless because of periodontal problem, were selected and were thoroughly planed to remove diseased cementum and divided into root-planed and citric acid-treated groups. After all of the teeth were longitudinally sectioned, the sectioned teeth were cut again to use middle third of the roots for normal group and period ontally diseased portion for root-planed and citric acid treated group. In the cell attachment experiment, human cultured fibroblasts were seeded in each culture well in which contained prepared roots and incubated for 4, 12 and 24 hours at 37C, 5% COJair incubator. Fibroblast attachment was measured by cell counting at each time point. In the cell proliferation experiment, all of the specimens were moved into fresh culture wells after 12 hours of incubation and incubated for 48, 72 and 96 hours. Fibroblast attachment was measured as the same of above. The results were as follows 1. In the cell attachment experiment, there was no statistic ally significant difference among the groups while the greater amount of cell attachment was found in the citric acid-treated group. (P>0.05) 2. In the cell proliferation experiment, there were statistically significant differences between the rootplaned and the citric acid-treated group after 48 hours of incubation and between the normal and the citric acid-treated group after 72, 96 hours of incubation (P0.05)
